Assessment of sanitary status of the important and imported citrus varieties against stubborn diseaseý.

Citrus stubborn disease is caused by the helical mollicute Spiroplasma citri. The most typical symptoms of stubborn disease can include chlorotic patterns on citrus leaves and small, lopsided fruit. This pathogen can be transmitted through infected grafts and by several species of leafhoppers. In the present study, attempts were made to detect S. citri in plant by indirect ELISA and Indicator Plant. We used Agdia polyclonal antibody for ElISA and Madam Vinous cultivar had been considered as indicator plant. Preparations of spiroplasma antigens partially purified from two infected citrus cultivars (Valencia and Salustiana) were collected from Ramsar Research institute and khorramabad station respectively. On the basis of indicator plant, results showed that both of cultivars are infected to S. citri. Symptoms to be observed were included: slow growth, short internodes between leaves, and mottled leaves. But we could not able to detect S. citri in these cultivars by ELISA. Therefore, The ELISA test are not sensitive enough to reliably detect S. citri in infected plant or Agdia polyclonal antibody is not suitable for detection of local spiroplasma strains.
