Phosphate Uptake 1 (PUP1): applications of gene-specific markers for breeding and evolution studies

Rice has a vast genetic diversity as a result of a long history of cultivation and domestication has made it an excellent model in several genetic studies for crop improvement. Phosphorus (P) is the second most important inorganic plant nutrient that is a vital component of key cell biological processes. However, phosphorus in the soil is limited or it is often present in forms that are unavailable to plants. Phosphorus deficiency has been recognized as a major abiotic stress limiting growth and productivity of rice throughout the world. Discovery of the major QTL for phosphorus update, Pup1 has been an important breakthrough in the development of phosphorus deficiency tolerant varieties. Based on the PUP1 sequence, gene-based markers were designed, validated, and used to asses the PUP1 haplotype in diverse rice germplasm to identify recipient parents for PUP 1 introgression and for phylogenetic analysis. PUP 1 introgression lines are being developed through marker-assisted back crossing (MABC) using PUP 1-foreground markers in conjunction with SNP-markers distributed throughout the genome. The development of PUP 1 introgression lines of two irrigated varieties (IR64, IR74) is at an advanced stage (BC2F4) and screening under P stress is ongoing. In addition to that, a phylogenetic study of the PUP 1 region has been initiated to understand its evolution in Oryza species. Selected Oryza sativa (L.) varieties and several wild relatives (AA. HHKK) were used. Prelimenary data showed that putatives kinase (PupK46) might have been conserved from O. rufipogon, O. glaberrima, and their ancestors.
