Studies on stability of exogenous gene in transgenic apples (Malus domestica Borkh.) | 外源基因在苹果转基因植株的稳定性研究

Китайский. 以继代培养13年的转豇豆胰蛋白酶抑制剂(CpTI)基因和新霉素磷酸转移酶(nptⅡ)基因苹果嘎拉、王林、乔纳金和富士组培苗及田间定植6~8年的转基因植株为试材,应用卡那霉素筛选和PCR扩增分析对外源基因的稳定性进行研究。结果表明,在57个苹果转基因株系组培苗中均可检测出CpTI特异基因片断;转化株系在添加50mg/L卡那霉素的培养基中生长正常。田间株系PCR检测结果表明,转入的外源CpTI基因在所测株系的叶片、花粉及大部分根系中稳定存在,有极少数株系的根系中未检测到特异表达条带。结果说明外源基因可以在转基因植株中稳定存在,但在不定根发生过程中有可能会丢失。

The stability of exogenous CpTI gene and nptⅡ gene in transgenic lines of Gala, Orin, Jonagold, and Fuji apples that either subcultured in vitro for 13 years or planted in field for 6 to 8 years was evaluated via kanamycin resistance test and PCR analysis. The results showed that the characteristic band of CpTI gene could be found in all 57 lines subcultured in vitro, and the plantlets could grow normally on the medium supplemented with 50 mg/L kanamycin. For the plants cultivated in field, the characteristic band of CpTI gene could be found in all leaf and pollen samples and the most of the root samples. A few root samples showed negative results. It indicated that the exogenous gene could remain in transgenic lines stably with an occasionally exception of in adventitious roots.
