Identification of natural vector(s) of Eggplant mottled dwarf virus in potato fields

Potato is one of main and strategic crops in Iran. Viral diseases are prevalent in this crop because of its vegetative propagation. Eggplant mottled dwarf virus (EMDV) is a plant rhabdovirus belongs to Nucleorhabdovirus genus. EMDV has a wide host range and cause serious damage to potato crop. In this project, for identifying of natural vector(s) of EMDV, several insect and mite samples were evaluated for their ability in transmission of EMDV. During 2001 till 2005, insect samples were collected from potato fields showing typical EMDV symptoms in Tehran, Esfahan, Chahar-Mahalva- Bakhtiary and Khorasan provinces. Studies on determining possible soil-born vectors, showed no soil-born vectors for EMDV, in these fields. The insect samples were reared on healthy suitable hosts and also potato and eggplant plants. These insects were then fed on infected potato and eggplant plants and then transferred on to healthy potatoes and eggplants. These plants were then maintained in greenhouse for symptom evaluation and then were tested by ELISA for EMDV. Among the insect species tested for transmission only Agallia vorobjevi can transmit EMDV from infected plants to healthy potatoes. After 3 to 4 weeks post inoculation, potato plants showed top necrosis and also eggplants showed vein yellowing. These plants and also Agallia vorobjevi vectors had positive reaction with specific EMDV antibodies in ELISA tests. These results confirm that Agallia vorobjevi is an insect vector for EMDV.
