Comparison of C.oedematiens culture in fermenter and Bottle

C.novyi (C. oedematiens) type B is causative agent of Black Disease in sheep and goat. Up to now C.novyi culture has been established in bottles. In this study we have tried to characterize and establish culture of microorganism in experimental fermenter. Although no publication has been so far published regarding the C.novyi culture, we tried to adapt bacteria with condition of fermenter and to get the best yield. So the media were prepared in fermenter and after a few tests we found sterilization temperature at 121°C for 45 minutes is the most appropriated for 10 litters of culture in experimental fermenter. The media were inoculated with different amount of seed including 1%, 5% and 10% in separate experiments. During 72 hrs of incubation period six samples war taken within 12 hrs interval and were used for detection MLD, doing spectophotometry and the other tests for evaluating exotoxins which release during growth and also for counting the number of bacteria in culture. We could record the pH variation in culture during 72 hrs incubation period for the first time in this research in which every (6 to 8) hrs it decreased 0.15 to 0.2. In some cases we adjusted pH at 7.2 again by adding NaOH 20%. When we had both inoculated medium by 5% seed and adjusted pH 7.2 during growth, we could get the most MLD, wherease we had not considered pH adjusting the MLD was obtained in fermenter as the same rate in bottle. Because of forming streptobacillary forms in fermenter we could not get significant result by Dark Field microscopy. In spectophotometry experiments we found interestingly OD went down 12 hrs after inoculating and in some cases after 24 hrs. Although it has been not mentioned by any references, we think decreasing OD causes by exotoxin effects on the microorganisms. The results which we found in this study will enable us to culture bacteria in big fermenters for mass production of vaccine.
