Introgression of genetic elements of non-tuberous type of Solanum etuberosum in potato by somatic hybridization means

Русский. Цель настоящей работы: использование молекулярного маркирования для анализа интрогрессии генетических элементов неклубненосных видов в соматические гибриды и их половые поколения. По итогам ПЦР-маркирования в половых поколениях соматических гибридов трех комбинаций нами выделено 27 генотипов с маркером COS4 на генетические элементы неклубненосного вида S. etuberosum, в том числе 17 для комбинации 2D, 8 - 4D, 2 - Р. Интрогрессия генетических элементов неклубненосных видов Solanum в геном культурного картофеля S. tuberosum посредством соматической гибридизации подтверждена SCAR-маркером COS4 на ген COSII_Atgl4320 из S. etuberosum. Представлена характеристика комбинаций с присутствием в родословной одного из родителей соматического гибрида с неклубненосными видами по наличию ПЦР-маркера COS4. Положительный ответ с ДНК-маркером COS4 получен для МВГ2С Н78-1, Н79-1, Н83-6, представляющих пятое половое поколение соматического гибрида 2D-265-2 - максимальная удаленность от вида S. etuberosum в нашем эксперименте. Полученные результаты свидетельствуют о реальной интрогрессии генетического материала неклубненосного вида S. etuberosum в геном культурного картофеля посредством соматической гибридизации.

Английский. The aim of this work is the use of molecular marking for the analysis of introgression of genetic elements of non-tuberous species in somatic hybrids and their sexual generation. As a result of PCR labeling in sexual generations of somatic hybrids of three combinations, we have allocated 27 genotypes with COS4 marker on genetic elements non-tuberous species S. etuberosum, including 17 for the combination of 2D, 8 - 4D, 2 - R. Introgression of non-tuberous genetic elements of Solanum types in cultivated potato genome of S. tuberosum by somatic hybridization was validated with SCAR-marker COS4 for COSII_Atgl4320 gene of S. etuberosum. There was presented a characteristic of combinations with the presence in progeny of one of the parents with somatic hybrid with non-tuberous species by the presence of a PCR marker COS4. A positive response with COS4 DNA marker is obtained for MVG2S N78-1, N79-1, N83-6, representing the fifth generation of sexual somatic hybrid 2D-265-2, which is in the maximum distance from the species S. etuberosum in the experiment. The obtained results indicate a real introgression of genetic material non-tuberous species S. etuberosum into the genome of cultivated potato by somatic hybridization.
