Optimization of polygalacturonase synthesis by aspergillus niger cntp 5037 in solid - state fermentation

Polygalacturonase has many applications in the food industry, textile industry, waste water treatment, production of animal feed. Besides, polygalacturonase also be used to produce pectic oligosaccharides (POS), known as a new prebiotic. Polygalacturonase is produced from various sources but mainly from A. niger in solid-state fermentation. To study the biosynthesis of polygalacturonase and find the optimum medium, Box-benken design was performed to investigate the effect of the moisture content, concentration of pectin, temperture and substrate thickness. Optimal conditions for the strain A .niger CNTP 5037 polygalacturonase biosynthesis include: moisture content 64%, pectin concentration 14%, temperature 27 °C, substrate thickness 17 mm. Using the optimized medium and addition ratio 2.106 spores/g with a 72-hour incubation period, the maximal activity of polygalacturonase production was found to be 25.02 U/g.
