Callus induction in VU-2 aym (Dioscorea alata L.) microtuber explants as affected by different levels of 2, 4-D

Conventional hybridization in yam (Dioscorea alata L.) is not possible since existing genotypes do not produce flowers under Philippine conditions. Production of genetic variants, however, has been reported through tissue culture techniques. This research was done to explore the use of in vitro microtubers as explants for callus induction (an initial step towards variability creation) in yam. In vitro-produced microtubers were cut into sections measuring about 1x8x8 mm (h x l x w) and inoculated in modified MS medium supplemented with different levels of 2, 4-D (0, 0.1, 0.2, 0.5, 1.0 and 2.0 mg/l). Results revealed that all cultures in 2, 4-D-supplemented media produced 100% callus formation except in 0.2 mg/L with only 95%. The earliest to produce callus tissues were those in medium with 1.0 mg/L 2, 4-D. Increasing the concentration further to 2 mg/L did not enhance callusing but rather caused delayed callus formation. In addition, cultures in medium with 0.2 mg/L produced the biggest callus tissues in terms of thickness, length and width from 2nd the 5th week of incubation.
