Chemical mutagenesis of Ganoderma lucidum for hyper production of monomeric manganese-dependent peroxidase (MnP)

One of the essential enzymes produced by White rot fungi (WRF) is Manganese Peroxidase engenders strong oxidants (combusts bodywork of lignin). MnP acts as generating and transferring oxidants, oxidizes (increase oxidation number of Mn2+ to Mn3+) the organic molecules. Light amber PDA imparts solid medium for sterling and uncontaminated culture of Ganoderma lucidum (GL). In the present research work, hyper-producing native and mutant strain of G. lucidum spawned by chemical mutagenesis with the help of chemicals Ethidium Bromide (EB) and Ethyl Methyl Sulphonate (EMS) will be studied. Mutants of Ganoderma lucidum will be withdrawn by screening methods. To measure the duration of growth and MnP production from dissimilar mutants will be the criterion to elect superlative mutant strain. MnP secreted by superlative mutant strain will go through optimization methodology comprising parameters (like effect of pH, temperature, carbon to nitrogen ratio, inoculum size, time of incubation, moisture level for enhanced enzyme mass production and low molecular mass mediators). Response Surface Methodology will be used for the optimization of factors under study.
