New strains of nodule bacteria, isolated from soils of Magadan region, and their physiological and cultural properties | Новые штаммы ризобий, выделенные из почв Магаданской области, и их физиолого-культуральные свойства

In order to study in detain Far Eastern natural populations of nodule bacteria, soil samples were obtained from under legume crops (chickling vetch, wild vetch) from Magadan Research Agricultural Institute in 2013. In 2014 under laboratory conditions seeds of soybean, and cowpea were sown in the received soil samples. In the flowering phase (June 2014), the root system of the grown leguminous plants was inspected. Small nodules were found only on the roots of cowpea of two species (Vigna radiata and V. unguiculata). Twenty strains of nodule bacteria were isolated to a pure culture from these nodules. All the isolated strains gave an abundant or good growth of the bacterial line on the agar mineral-plant medium (MPM) with mannitol. On the MPM with lactose these strains sharply reduced the intensity of growth. Only two bacterial cultures (Md-0, Md-13) grew on the meat-and-peptone agar (control medium). On the MPM with mannitol and bromthymol blue indicator all the studied strains had a heavy growth of the stroke with the release of metabolic products of the acidic or neutral character at the initial stage of growth. 50% of stains did not form nodules on the roots of soybean Garmonia, the virulence of the other strains of nodule bacteria was 10–30%. At the same time, all pure nodule cultures very actively formed nodules on the roots of cowpea (V. unguiculata). According to the preliminary results of small-scale field tests on meadow chernozem-like soils, the strains Md-16 and Md-7 increased the productivity of phytomass of soybean Eugenia by 4,2–7,5 c/ha (from 58.4 c/ha), and the yield of seeds – by 1.4–3.1 c/ha (from 25.4 c/ha). It is assumed that the isolated strains belong to the species Bradyrhizobium elkani.

С целью всестороннего изучения дальневосточных природных популяций ризобий в 2013 г. из Магаданского НИИ сельского хозяйства были получены образцы почвы из-под бобовых культур (чина, мышиный горошек). В 2014 г. в лабораторных условиях в эту почву посеяли семена сои и вигны. В фазе цветения культур (июнь 2014 г.) провели осмотр корневой системы растений. Мелкие клубеньки обнаружили только на вигне двух видов (V. radiata и V. unguiculata). Из этих клубеньков в чистую культуру выделили 20 штаммов ризобий. Все они давали обильный или хороший рост штриха бактериальной культуры на агаризованной минерально-растительной среде (МРС) с маннитом. На питательной МРС с лактозой эти штаммы заметно снизили интенсивность роста штриха. Только две культуры бактерий (Мд-0, Мд-13) дали рост штриха на мясо-пептонном агаре (контрольной среде). На МРС с маннитом и индикатором бромтимоловым синим все изученные штаммы давали обильный рост штриха с выделением продуктов метаболизма кислотного или нейтрального характера на начальном этапе роста. Не образовывали клубеньки на корнях сои сорта Гармония 50% штаммов, вирулентность остальных штаммов ризобий составляла 10-30%. В то же время все чистые культуры ризобий очень активно формировали клубеньки на корнях вигны (V. unguiculata). По предварительным результатам полевых испытаний в мелкоделяночных опытах на луговых чернозёмовидных почвах, штаммы Мд-16 и Мд-7 повысили продуктивность фитомассы сои сорта Евгения на 4,2-7,5 ц/га (c 58,4 ц/га), урожайность семян – на 1,4-3,1 ц/га (с 25,4 ц/га). Предполагаем, что выделенные штаммы относятся к виду Bradyrhizobium elkani.
