Flavonoids, as plant-derived natural compounds, have a wide range of health-promoting effects such as antioxidant, anti-inflammatory, anticancer and others; thus, worldwide interest towards its research is increasing. A local medicinal plant rich in flavonoids is Kaempferia galangal L. Its antioxidant, anticancer and wound healing activities are attributed to its flavoniod content, mainly of quertin, kaempferol and apigenin. Accurate identification of this plant is important. In this study, K. galanga was differentiated from its closely related species, K. pulchura, through morpho-anatomical characterization, molecular diversity analyses (SSRs, psbA-trnH and petA-psbJ-psbL) and flavonoid profiling. The analyses were robust enough to authenticate local K. galanga. Within K. galanga accessions, intraspecific variation was noted based on combined psbA-trnH and partial petA-psbJ-psbL sequences. Presence and amounts of the three dominant flavonoids varied in the different organs of K. galanga as revealed by TLC and HPLC. Callus cultures of the plant produced and sequestered flavonoids and differential accumulation of these compounds was elicited by yeast extract. Authenticated local K. galanga and its callus cultures, thus can be good sources of health promoting flavonoids.
