Components of the dynamics of the interaction between maize and lepidoptera stem borers | Composantes de la dynamique de l'interaction entre le maïs et les insectes lépidoptères foreurs de tige

Французский. Les dégâts causés par les larves des lépidoptères foreurs de tige comme la pyrale du maïs Ostrinia nubilalis constituent l’une des contraintes majeures à la production de maïs. La sélection de variétés résistantes ou tolérantes à ces ravageurs est l’un des moyens de lutte contre les infestations naturelles. Pour identifier ces variétés, j’ai mis au point une méthode originale permettant d’évaluer, à grande échelle et de façon non destructive, l’appétence des larves de pyrale pour des feuilles de maïs. La vitesse de consommation des larves, mesurée au laboratoire à partir de disques foliaires prélevés sur des plantes vivantes, est utilisée comme un proxy du niveau de défense des plantes. J’ai démontré que les larves de pyrale ont des comportements variables lorsqu’elles sont mises en présence d’un disque foliaire de maïs, et qu’il est possible d’établir une typologie de leurs comportements alimentaires, avec une graduation allant de la consommation rapide et entière du disque à l’absence total de consommation. Chaque variété peut ainsi être caractérisés de façon reproductible par la fréquence de chacun des types de comportement alimentaire. Après avoir validé ce dispositif par l’étude des variations de comportements alimentaires des larves en présence de différentes concentrations de substances antiappétentes, je l’ai utilisé sur différents jeux de données. Tout d’abord, j’ai comparé des génotypes précoces et tardifs issus d’une expérience de sélection divergente pour la précocité de floraison. Je montre que les génotypes les plus contrastés pour la date de floraison, descendants par sélection et autofécondation de la lignée de maïs MBS, présentent des différences importantes à la fois pour l’appétence des larves pour les feuilles, pour la prévalence des larves sur les plantes au champ, et pour la qualité des parois cellulaires. Les génotypes MBS précoces sont plus appétents, plus attaquée et plus tendres que leurs homologues tardifs. Ces résultats confortent l’existence chez le maïs, d’un compromis entre croissance et défense. Enfin, le test de consommation a été appliqué à panel de 18 lignées de maïs déjà décrites pour un ensemble de traits liés au métabolisme et à la physiologie des plantes. L’étude confirme l’existence, au sein du panel, d’une grande variabilité entre lignées pour l’appétence des larves de pyrale et permet d’identifier des caractéristiques métaboliques des lignées corrélées aux variations des types de comportements alimentaires. Dans l’ensemble, cette étude ouvre la voie vers de nouvelles approches pour mieux comprendre les interactions plantes-insectes ravageurs.

Английский. Damages caused by maize stem borers like the corn borer Ostrinia nubilalis can engender considerable yield reductions. A way to reduce the damages is to select resistant or tolerant maize varieties. I set-up an original high-throughput and non-destructive method to screen maize varieties for the feeding of young insect larvae. It consists in monitoring the feeding of single larvae on fresh maize leaf disks, and using the consumption speed as a proxy for the level of plant defenses. I showed that larvae exhibit variable feeding behaviors, and developed a statistical method to establish a typology of feeding behaviors, with a graduation between the fast and full consumption of the leaf disk and the absence of consumption. Each maize variety can be reproductively characterized by the frequency of each feeding behavior. The method was validated by studying the variations of feeding behaviors in response to different concentrations of a known antifeedant molecule, and applied on different datasets. First, I compared early and late maize genotypes resulting from a divergent selection experiment for flowering time. I showed that the genotypes that differed the most for flowering time and that descended, through selection and selfing, from the inbred line MBS, also showed important differences concerning both feeding behaviors, larvae prevalence in the field, and the quality of plant cell walls. Early MBS genotypes are consumed faster in the feeding test, show a higher larvae prevalence in the field, and exhibit softer green parts than late genotypes from the same ancestor line. Those results strongly suggest the existence of a trade-off between growth and defense. Finally, the consumption test was applied to a diversity panel of 18 maize inbred lines, already described for series of traits linked to plant physiology and metabolism. I showed that the panel lines exhibited a large genetic variability for the feeding behavior of corn-borer larvae and identified metabolic characteristics of the inbred lines that correlate with the variation of feeding behaviors. Altogether, those results open the way for a better-understanding of plant-insect relationships.
