Cellular location of polyphenol oxidase in Metroxylon sagu

The location of polyphenol oxidase (PPO) in cells of sago pith tissues was examined histochemically and viwed under light microscope. In pith tissues collected from 3-4 years sago log a large number of vascular bundles and vacuoles occupied most of the cells. While in young sago pith (2-3 months) the vacuoles occupied far less of the cytoplasm and dense pigmentations can be seen in some of the cytoplasmatic organelles. Using catechol for histochemical staining of PPO, dense deposition of polyphenol oxidase reaction product is readily visualized within the xylem cell walls and the parenchyma cell walls. These results reveal that sago ptih browning is due to highly localized oxidative activity mediated by polyphenol oxidase.
