Effect of extraction methods for analysis fatty acid in the milk fat

The aim of this work to compare the ultracentrifugation method with other extraction methods, which are used for determination of fatty acids (FA) in milk fat, depending on their time demands and safety while maintaining their quantitative parameters. The content of FA was determined in pool samples (n = 12) of milk from Holstein cows from a farm in the Olomouc region. In this work we present the results of analysis of FA in the milk fat, which we analysed by 5 different extracion methods. We used partition extraction method, Rös-Gottlieb extraction, cold sample centrifugation, ultracentrifugation method and lyophilization method. The obtained fat extract samples were subsequently derivatized to fatty acid methyl esters using the basis esterification method. Then we analysed the total FA profile on the gas chromatograph. After analyses of FA we divided FA into four groups: saturated FA (SFA), mono-unsaturated FA (MUFA), poly-unsaturated FA (PUFA) and trans-izomer unsaturated FA (TFA). By evaluating the results we found significant differences between the ultracentrifugation extraction method and the partition extraction at the significance level P= 0.001 in the case of SFA group determination. At the same time, however, it can be stated that the results can be considered identical for all other extraction procedures.
