Express methods for post-vaccinal diagnosis of brucellosis | Экспресс-методы поствакцинальной диагностики бруцеллеза

Русский. Отработана технология экспресс-методов диагностики на фоне специфической профилактики бруцеллеза крупного рогатого скота. Для исследования были использованы: роз-бенгаловая проба (РБП) как наиболее чувствительный метод и реакция иммунодиффузии (РИД) с О-ПС антигеном как дифференциальный тест при оздоровлении стад крупного рогатого скота. Для выявления скрытого течения бруцеллеза, используя метод провокации, исследовали сыворотку крови коров, привитых вакциной из шт. B. abortus 82 в короткие сроки (через 21 день – до 3 месяцев) после иммунизации на фермах с разной эпизоотической ситуацией по бруцеллезу. В стадах неиммунного поголовья как рекогносцировочный тест при установлении предварительного диагноза на бруцеллез используется РБП. Результаты оценки роз-бенгал пробы учитывают только как "положительные" и " отрицательные". Установлено, что поствакцинальные реакции РБП, оцениваемой в крестах, полностью совпадают с показателями реакции агглютинации (РА). У здоровых животных, привитых вакциной из шт. 82, показатели реакций невысокие, в отличие от больных бруцеллезом животных, подтверждается это в экспериментах с искусственным заражением интактных и иммунизированных вакциной шт. 82 коров из неблагополучных по бруцеллезу пунктов. Показатели РБП "+++" и "++++" соответствуют РА в титрах 200 МЕ и выше. Более того, высокие показатели РБП поглощают показатели реакции связывания комплемента (РСК) в разведении 1 : 10 и выше, а также и РИД. Этот факт дал возможность использования РБП взамен классических реакций – РА и РСК. Применение РБП наиболее эффективно при выявлении латентно больных животных, в комплексе с реакцией иммунодиффузии с О-полисахарадным антигеном в короткие сроки после иммунизации вакциной из шт. B. abortus 82.

Английский. The technology of express diagnostic methods against the background of specific prophylaxis of brucellosis in cattle has been developed. The Rose Bengal Test (RBT) as the most sensitive method and immunodiffusion test (IDT) with O-PS antigen as a differential test for the improvement of cattle herds were used for the study. Using the provocation method for detection the latent course of brucellosis there was studied the blood serum of cows vaccinated with the B abortus 82 strain vaccine in a short time (in 21 days – up to 3 months) after immunization on farms with different brucellosis epizootic situation. According to the instruction of RBT, it is used in herds of non-immune livestock as a reconnaissance test when establishing a preliminary diagnosis for brucellosis. Therefore, the evaluation results of the Rose Bengal Test of the sample are taken into account only as "positive" and "negative". It was found that the post-vaccination reactions of RBT, evaluated in crosses, completely coincide with the indicators of the agglutination test (AT). The experiments with artificial infection of intact and immunized with a strain 82 cows from brucellosis-prone areas confirmed that in healthy animals vaccinated with the vaccine from strain 82, the reaction rates are low, in contrast to animals with brucellosis. RBT indicators "+++" and "++++" correspond to AT in titers of 200 IU and above. Moreover, high levels of RBT absorb the indicators of the complement-fixation test (CFT) at a dilution of 1 : 10 and above, as well as IDT. This fact made it possible to use RBT instead of the classical tests such as AT and CFT. The use of RBT proved to be the most effective when identifying latently ill animals, in combination with the IDT with the O-PS antigen in a short time after immunization with the vaccine from strain B. abortus 82.
