Cloning of the gene for the enzyme phytase from Aspergillus niger [in maize kernels, in order to reduce phosphorus excretion]

Английский. Phosphorus is an essential element for the growth of all organisms. An insoluble form of phosphorus occurs as storage source in virtually all substances originating from plants. This phosphorus, myo-inositolhesakis-phosphate or phytate or phytic acid, is solubilized by the enzyme phytase (EC 3.1.3.8). Phytic acid is considered to be an antinutritional factor since it chelates minerals and may also react with proteins, thereby decreasing the bioavailability of proteins and nutritionally important minerals. The excretion of phytate phosphorus in manure has also consequences on environment; this is due, partially, to the accumulation of phosphate from manure in surface waters which cause eutrophication. The enzymes, produced by microorganisms and plants, that catalyze the conversion of phytate to inositol and inorganic phosphorus are known as phytases. итальянский. Il fosforo e' un elemento essenziale per la crescita di tutti gli organismi. Una forma insolubile di fosforo e' presente in buona parte delle sostanze di origine vegetale. Questo fosforo, mioinositolesachis-fosfato o fitato o acido fitico, viene solubilizzato dall'enzima fitasi (EC 3.1.3.8). L'acido fitico viene considerato un fattore antinutrizionale, dal momento che e' in grado di chelare gli elementi minerali e interagisce con le proteine determinando un decremento della biodisponibilita' delle proteine e di importanti elementi minerali. La eliminazione del fosforo fitinico con le deiezioni animali ha ulteriori conseguenze negative sull'ambiente; questo a causa, in parte, dell'accumulo del fosforo nelle acque superficiali, con conseguente eutrofizzazione. Gli enzimi, prodotti da microorganismi e da piante, che catalizzano la conversione del fitato in inositolo e fosforo inorganico, sono conosciuti come fitasi. In questo lavoro viene descritto il clonaggio di un gene per l'enzima fitasi da Aspergillus niger
