Sections fed with sulforhodamine G were prepared by freeze-substitution and resin embedding, and studied by microspectrofluorometry. Tracer was found to move within cell walls in restricted pathways. The highest fluorescent intensity was found around the border of mestome-sheath and parenchyma-sheath; this is shown to be due to osmotic separation, and given as evidence of a major symplastic water movement.
