In vitro culture: Environmental conditions and plantlet growth as affected by vessel and stopper types

In conventional micropropagation cultivation media usually contain saccharides, mainly saccharose, as the sources of carbon and energy. However, it has been described recently that plantlets from photosynthetic apparatus enabling photoautotrophy in suitable environments, i.e. under sufficient irradiance and CO2 content. Plantlet irradiance, CO2 influx and water vapour efflux were affected by vessel type and vessel closure. High transparency of vessel covers for light or lateral lighting and high permeability of vessel stoppers for CO2 or higher CO2 concentration in ambient air increased photosynthetic activity and plantlet growth. In addition higher permeability of covers decreased negative effect of ethylene accumulation on plantlet morphology and dry mass accumulation. Fully autotrophic cultivation of plantlets on medium without saccharose is a prospective type of future technology.
