[Polyclonal antibody use on filtrate of surface washing mycelium culture to develop early detection of Ganoderma sp. infection]

Английский. Butt rot incited by Ganoderma sp. is one of the most important diseases of oil palm. Forty percent of the stands at productive stage are usually damaged due to the disease. The development of early detection technique using Enzyme-Linked Immunosorbent Assay based on polyclonal antibody (ELISA PcAb) has been conducted. A polyclonal antibody employed in the study was prepared by immunizing Bal b/c mice using antigen obtained from filtrate of surface washing mycelium culture of isolate TK-1. The ELISA developed in the present study showed to be able to detect 0.0012 mg antigen per ml filtrate of a 14 day mycellium culture. A considerable cross-reaction was experienced in 16 of 18 antigens of different isolates prepared from filtrate of surface-washing mycelium of in vitro cultures. All antigens prepared by washing of tissue and spores except spore of isolate TK-1 could not be detected using of the ELISA-PcAb. Attemps to increase specificity of the detection method have been initiated by the production of monoclonal antibody and the use of antigen obtained from intact tissue of a fruiting body

индонезийский. Penyakit busuk pangkal batang yang disebabkan oleh Ganoderma sp. merupakan penyakit paling penting pada tanaman kelapa sawit. Kematian yang diakibatkan mencapai 40 persen tegakan pada saat tanaman mencapai umur produktif. Pengembangan teknik deteksi penyakit secara ini berdasarkan metode Enzyme-Linked Immunosorbent Assay untuk melacak keberadaaan material antigenik Ganoderma sp. menggunakan antibodi poliklonal (ELISA-PcAb) sedang dikaji. Antibodi poliklonal anti Ganoderma sp. dibuat dengan cara imunisasi mencit Balb/c dengan filtrat pencucian kultur miselium isolat TK-1. Hal ini dilakukan sebagai kegiatan penelitian pendahuluan dalam produksi dan karakterisasi antibodi monoklonal anti Ganoderma sp. Penggunaan serum poliklonal segar yang diperoleh dalam ELISA-PcAb mampu mendeteksi 0,0012 mg antigen/ml filtrat dari kultur umur 14 hari. Akan tetapi dengan menggunakan ELISA-PcAb tersebut diketahui bahwa reaksi silang dapat terjadi pada 16 dari 18 isolat Ganoderma sp. yang diuji. Semua antigen yang berasal dari jaringan tubuh buah dan spora, kecuali spora dari isolat TK-1 tidak dapat dideteksi dengan ELISA-PcAb tersebut. Untuk meningkatkan spesifitas pendeteksian kajian mengenai kegunaan antibodi monoklonal dan penggunaan antigen dari jaringan tubuh buah yang intak dengan tanaman sedang dilakukan
