Hatching in the potato cyst nematode Globodera rostochiensis indicating diapause as a cause of variability in emergence [Solanum tuberosum]

Over a period of 12 months new cysts (extracted soon after maturity on host roots) and old cysts of Globodera rostochiensis (stored for one calendar year outdoors in a gravel plunge) were hatched in potato root diffusate. Diapause was evident in new cysts in early autumn, winter, spring and mid-summer but was absent in late autumn and summer. By contrast when old cysts were hatched, emergence was about 90% in all periods. It is suggested that diapause persists in new cysts, but is overcome when cysts are stored for 12 months in the soil outdoors. A comparison of infectivity of juveniles obtained in the periods when eggs have overcome their diapause failed to show any differences. Also when the number of eggs in new and old cysts was compared, the number of eggs in old cysts exceeded the number of eggs in new cysts. It is suggested that hatching in old cysts is due to absence of diapause. The presence of large numbers of eggs in old cysts even after 12 months of storage outdoors in the soil failed to support the theories of hatching, microorganism induced hatching or persistence of hatching factors in the soil

[Nel corso di un periodo di 12 mesi cisti nuove (estratte subito dopo la maturita' dalle radici di patata) e cisti vecchie (conservate per un anno solare all'esterno in ghiaia situata nel terreno) sono state fatte schiudere in un liquido di diffusione di radici di patata. La diapausa era evidente nelle cisti nuove all'inizio dell'autunno, in inverno e in primavera e a meta' dell'estate, ma non alla fine dell'autunno e dell'estate. Al contrario, quando si facevano schiudere le cisti vecchie, l'emergenza risultava pari a circa il 90% in tutti i periodi dell'anno. Cio' fa pensare che la diapausa persista nelle cisti nuove, ma che venga superata quando le cisti sono conservate nel terreno, all'aperto, per 12 mesi. Il confronto sull'infettivita' delle larve ottenute nei periodi in cui le cisti avevano superato la diapausa non ha consentito di evidenziare alcuna differenza. Inoltre, quando si confrontava il numero di uova nelle cisti nuove e in quelle vecchie, queste ultime ne presentavano un numero maggiore. Si ritiene che la schiusa nelle cisti vecchie sia dovuta all'assenza di diapausa.
