Micropropagation of Cedrus sp (preliminary results)

Excised shoot tips two year old seedlings of Cedrus atlantica were cultured in SH medium supplemented with several combination of BA and NAA. At 0.5 mg/1 BA the number of shoots was greatest. All the formed buds were axillary and no adventitious shoot formation occurred. At 25o C, the buds formed were healthier than at 27o C, whereas a day/night alternating temperature 23-24o C/22-23o C) was better than a constant temperature of 27o C for the elongation of the shoots. Shoot elongation was carried out in a hormone free medium for four weeks and the average increase ranged between 0.4 and 0.95 cm depending on the treatment. Subsequent transfer of the explants into a full strength medium resulted in a more rejuvenated explants and an average of 10 new axillary buds were formed. The spraying of the mother explants with BA didn't affect the number of shoots formed whereas the date of collection of the shoot tips was more determinant. The response of the shoot tips and the number of buds and shoots were better in July and September than in August, coinciding with the growth period of plants. Green and friable callus was formed at 0.05 mg/1 NAA at the base of the explants. Rooting from this callus failed in both peat: prelite and agar solidified medium supplemented with IBA and NAA.
