Expression of ascorbic acid oxidase isoenzymes during development and ripening of melon (Cucumis melo) fruit

Activity, protein and isoenzymic profiles of ascorbic acid oxidase (AAO) were studied during development and ripening of melon (Cucumis melo L. var. reticalatus) fruit and in response to tissue and species specificity. During fruit development, the activity and protein content of AAO were mainly expressed in ovaries and preclimatic mature fruits. Moreover, AAO accumulated actively in the growing tissues especially in the apex. Cucumber ovaries (Cucumis sativus) are the richest source of AAO activity among all the species of Cucurbitaceae family. Ascorbic acid oxidase extracted from melon ovaries was fractioned by native isoelectric focusing (pH range 3 to 10) into at least 12 isoenzymes. Intercellular washing fluid revealed that 5 isoenzymes were in the particulate fraction of the extract and 7 isoenzymes were present in the soluble fraction of the extract
