Constitutive diamine oxidase (DAO) activity has been investigated in internodes and leaves of different node levels obtained from Vicia faba cultivars susceptible and resistant to the antrachnose. This activity has also been investigated in stems and leaves of a whole plant and in detached leaves, untreated or artificially infected with different conidia concentrations of one of the most virulent Ascochyta fabae isolates on V. faba, obtained from the susceptible cultivar Lunga delle Cascine. DAO activity is higher in the leaves than in the stems, it decreases acropetally along the plant and it was not detected any increase of this activity as response to the inoculation. V. faba internodes and leaves, wounded by mechanical injury, was the biological system used to elucidate the possible role of DAO in the response to external stimuli. Time courses of DAO activity was analysed during the wound healing of the third internode and leaf of the cultivar Lunga delle Cascine; this activity was strongly enhanced by mechanical injury five days after wounding and apparent histochemical activity of diamine oxidase was detected in the lignosuberized barriers

[E' stata studiata l'attivita' della diamina ossidasi (DAO) in internodi e foglie di differenti livelli di nodi prelevati da cultivar di Vicia faba suscettibili e resistenti all'antracnosi. Questa attivita' e' stata pure studiata in fusti e foglie di una pianta intera e in foglie staccate, non trattati o infettati artificialmente con concentrazioni diverse di conidi di uno degli isolati piu' virulenti di Ascochyta fabae su V. faba, ottenuti dalla cultivar suscettibile Lunga delle Cascine. L'attivita' di DAO e' piu' alta nelle foglie rispetto ai fusti, decresce in direzione acropeta lungo la pianta e non e' stato rilevato alcun aumento di questa attivita' come risposta all'inoculazione. La produzione di ferite meccaniche in internodi e foglie di V. faba era il sistema biologico utilizzato per mettere in luce il possibile ruolo di DAO nella risposta agli stimoli esterni. L'andamento nel tempo dell'attivita' di DAO e' stato analizzato durante la cicatrizzazione delle ferite praticate nel terzo internodo e nella foglia della cultivar Lunga delle Cascine; questa attivita' era fortemente incrementata dal danno meccanico cinque giorni dopo che le ferite erano state praticate e l'attivita' istochimica apparente della diamina ossidasi era riscontrata nelle barriere lignosuberificate]
