Recent possibilities for the diagnosis of early pregnancy and embryonic mortality

Transrectal ultrasonography was used at 3 to 4 day intervals between days 26 and 45 and between days 53 to 58 after AI to evaluate the time of spontaneous late embryonic/foetal mortality (LEM/FM). There were 11 of 138 cows (8.0%) with LEM/FM. Levels of progesterone, bovine pregnancy specific protein B (PSPB) and bovine pregnancy associated glycoprotein 1 (PAG1) were measured by radioimmunoassay in plasma samples withdrawn after each ultrasound diagnosis. Luteal regression was obviously not the cause of conceptual death. The PSPB and PAG1 concentrations started to decrease steadily and during the period under observation reached the cut-off values in 8 and 4 of 11 cows, respectively, which suggest a somewhat longer half-life for PAG1 than for PSPB. Two cows showed first a further small albeit temporary increase in their protein levels, before levels finally declined, suggesting that protein production by the binucleate cells was not yet disturbed at the time that LEM had occurred

[L'ultrasonografia transrettale e' stata utilizzata a intervalli di 3-4 giorni fra i giorni 26 e 45 e fra i giorni da 53 a 58 dopo la fecondazione artificiale per valutare il tempo della mortalita' embrionale/fetale tardiva spontanea (LEM/FM). Di 138 vacche, 11 (8,0%) sono risultate con LEM/FM. In campioni di plasma prelevati dopo ogni diagnosi effettuata con gli ultrasuoni sono stati misurati, mediante saggio radioimmunologico, i livelli di progesterone, di proteina B specifica della gravidanza bovina (PSPB) e di glicoproteina 1 associata alla gravidanza bovina (PAG1). La regressione luteale non era ovviamente la causa della morte nel concepimento. Le concentrazioni di PSPB e di PAG1 iniziavano a diminuire con continuita' e durante il periodo di osservazione raggiungevano i valori di soglia minima in 8 e 14, rispettivamente, delle 11 vacche, il che fa pensare a una semivita alquanto piu' lunga per PAG1 rispetto a PSPB. Due vacche hanno presentato dapprima un ulteriore leggero incremento, seppur temporaneo, nei loro livelli delle proteine, prima che gli stessi diminuissero definitivamente, facendo ipotizzare che la produzione delle proteine da parte delle cellule binucleate non fosse ancora influenzata al momento in cui si verificava il LEM]
