In vitro culture of olive cultivars (Olea europea L.), Kalamata and Koroneiki

The research scheme was carried out to study the effect of using single-node explants at different degree of juvenility, three kinds of media and different concentration of growth regulators in medium on growth and proliferation of aseptically cultured explants of olive in vitro. Single-node explants collected from succulent suckers were found to be more effective for proliferation than explants from vigorous shoots of cut-back trees or from vigorous shoots of adult tree. The juvenility degree of suckers was found to have a beneficial effect in increasing bud sprouting, mean axillary shoot length and total leaf number per shoot in the initation stage for cv. 'Koroneiki'. The best growth parameters were observed in the initation stage for 'Koroneiki' explants inoculated in OM plus 1 or 2 mg.1-1 IBA. There were not significant differences, among the three media tested (OM, K&H and WPM), on the growth parameters on the initiation stage for cv. 'Kalamata'
