Identification of a Triticum durum - Secale africanum amphiploid and its crossability with common wheat [Triticum aestivum L.]

An amphiploid between Triticum durum cv Cocorit and Secale africanum was evaluated by cytological observation, seed storage protein electrophoresis analysis and disease resistance surveys. Feulgen staining and Giemsa-C banding of somatic metaphases indicated that the nucleolus from S. africanum did not express in the amphiploid and a high percentage of aneuploid existed in the amphiploid population. APAGE showed that most gliadin of both parents was observed in the endosperm of the amphiploid with codominant expression. Inoculated by the stripe rust and powdery mildew isolates, the amphiploid did not express the resistance from S. africanum. In the crosses between the amphiploid and several wheat lines, the crossability of amphiploid as a female parent was high, and resulted in more hybrid plants compared with the reciprocal crosses. The different fertility of F1 plant of amphiploid x wheat seemed to depend on the genotypes of wheat parents involved. But the F1 plant from wheat line J-11 showed semi-fertility of female gamete, significantly higher than other crosses. It is likely that the factor(s) responsible for fertility-restored would exist in line J-11. [Un anfidiploide fra Triticum durum cv Cocorit e Secale africanum e' stato valutato mediante osservazione citologica, analisi elettroforetica delle proteine di riserva del seme e indagini sulla resistenza alle malattie. La colorazione di Feulgen e il bandeggio Giemsa-C delle metafasi somatiche indicavano che il nucleolo derivante da S. africanum non si esprimeva nell'anfidiploide e nella popolazione anfidiploide esisteva una percentuale elevata di aneuploidi. L'APAGE evidenziava che la maggior parte delle gliadine dei due parentali era presente nell'endosperma dell'anfidiploide con espressione codominante. Inoculato con isolati della ruggine striata e dell'oidio, l'anfidiploide non esprimeva la resistenza derivante da S. africanum. Negli incroci fra l'anfidiploide e numerose linee di frumento, l'incrociabilita' dell'anfidiploide come parentale femmina risultava elevata e si traduceva nella produzione di piu' piante ibride rispetto agli incroci reciproci. La fertilita' diversa della pianta F1 dell'incrocio anfidiploide x frumento sembrava dipendere dai genotipi dei parentali di frumento coinvolti. Ma la pianta F1 derivante dall'incrocio con la linea di frumento J-11 presentava una semi-fertilita' del gamete femminile, significativamente piu' elevata rispetto agli altri incroci. E' probabile che il (i) fattore(i) responsabile(i) per il ripristino della fertilita' esista(no) nella linea J-11.
