Pedigree analysis through ssr markers of Malus progenies obtained by crossing with apomictic species

Английский. The ability of flowering plants to propagate by seed is not necessarily linked to sexuality. Some angiosperms belonging to the families of Poaceae, Asteraceae, Rosaceae and Rutaceae commonly reproduce asexually through seed by a process called apomixis. Progenies derived from apomictic parents are genetic copies of the maternal genotype. As apomixis is not obliged into the genus Malus, we planned to carry out controlled crosses between completely no-parthenogenic and highly apomictic apple accessions, aiming at generating plant material resistant to apple proliferation. Three sexual (M9, P22 and Prima) and five apomictic selections of rootstocks were used in different combinations, including some reverse crossings. More than 1,600 seedlings were DNA typed using codominat SSR markers, in order to establish their pedigrees and start the characterization of new genetic resources for breeding of apple rootstocks. Results showed two main classes of allele combinations in the progenies: one identical to the mother genotype and the second having a third allele at each locus segregating from the male parent. Very few hybrids were observed when flowers from apomictic rootstocks were pollinated with the commercial M9 variety. The reverse crossings generated a higher percentage of recombinant seedlings, together with an unexpected number of self pollination events
Показать больше [+] Меньше [-]итальянский. La capacita' delle piante di propagarsi mediante seme non e' esclusivamente legata al fenomeno di riproduzione sessuale. Alcune angiosperme comunemente si riproducono mediante seme in modo asessuato, con un processo chiamato apomissia. Le progenie derivanti sono copie identiche del genotipo materno. Poiche' l'apomissia non e' obbligata all'interno del genere Malus, nel tentativo di produrre piante resistenti agli scopazzi del melo (AP), sono state impostate diverse combinazioni di incrocio tra tre varieta' di Malus x domestica (M9, P22 e Prima) e cinque selezioni di portinnesti apomittici. I 1.600 semenzali ottenuti sono stati tipizzati utilizzando i marcatori codominanti microsatelliti, allo scopo di stabilire il loro effettivo pedigree e caratterizzare nuove risorse genetiche per il miglioramento genetico del melo. I risultati indicano due principali classi di combinazioni alleliche nelle progenie: una identica al genotipo della madre e la seconda avente, oltre al genotipo della madre, un terzo allele derivante dalla segregazione, a ciascun locus, del genotipo paterno. Pochi ibridi sono stati osservati quando gli apomittici venivano impollinati con M9. Gli incroci reciproci hanno invece generato un'alta percentuale di semenzali ricombinanti, oltre a un'elevata presenza di eventi di autoimpollinazione inattesi
