Genetic characterization of the oriental fruit fly Bactrocera occipitalis (Bezzi) and B. philippinensis Drew and Hancock through molecular analysis and interspecific hybridization

Bactrocera philippinensis and B. occipitalis, are sympatric species in the Philippines that affect fruit products for local and export market. Samples of the two sympatric species, including their intermediates were subjected to molecular analysis and hybridization. Phylogenetic Bayesian inference on the fruit flies* ISI and period genes and in combination of the two genes were evaluated. The phylogenetic trees from period gene and /7SI gene show some clustering of the two species yet the intermediates did not form a separate cluster. Merging the two genes simultaneously in *BEAST, result show no distinct clusters for B. philippinensis and B. occipitalis were observed. Species delimitation test indicate that the two groups are not distinct. Hybrids were produced in a cross between female B. occipitalis and male B. philippinensis, but the reciprocal cross between the species did not produce any offspring. Backcrosses of the F, to either pure lines of B. philippinensis and B. occipitalis produce offspring. Hybridization of B. philippinensis and B. occipitalis show that medial line on tergite 4, transverse dark band on abdominal tergite 2 and lateral marks on tergite 4 are quantitative traits. The ground color of the frontal setae shows that the black foreground color is dominant over yellow.
