A commentary is given on a published human study on the dosing with an alpha-amylase inhibitor to inhibit starch digestion in the gastrointestinal tract, with comments also given on a complementary human double-blind crossover study using a starch blocker. In the 1st study, dosing a high-starch meal with alpha-amylase did not block starch digestion, as evidenced by fecal analysis for caloric content. In the complementary study, no effect on serum glucose, insulin, or breath hydrogen was obtained using a starch blocker. While the results of the former study did not result in excess caloric loss in the stool, the latter study indicated that this failure primarily was due to a lack of inhibitory effect on starch hydrolysis in the small intestine.
