Extraction and estimation of thymoquinone (a highly valued metabolite) from Nigella sativa L

The pharmacological activities of Nigella or Kalonji (Nigella sativa L.) are mostly attributed to its seed oil, of which thymoquinone is a significant bioactive component. A very limited standard formulations or thymoquinone-rich nigella extracts are currently in clinical use, most likely due to poor extraction and lower stability of the compound. In this study the extraction is done by ultrasonic extraction process and percent composition, active metabolites and the antioxidant action of Kalonji seed extract was estimated. For the quantification of thymoquinone, an HPLC method (C-18 column, 250 x 4.6 mm ID, with a particle size 5 μm and a flow rate of 1.50 mL min⁻¹) was followed in an isocratic flow system. The extract has yielded 5.820 ± 0.037%, and per cent composition of thymoquinone was 0.90%. Further the kalonji extract has reported the metabolites constituents’ viz., total phenolic compounds (31.850 ± 0.976 mg GAE g⁻¹ seed), flavonoid content of 7.310 ± 0.119 mg QE g⁻¹ of seed and has reported 76.180 ± 1.780% antioxidant action when measured using the DPPH free radical test. The HPLC analysis reveals that the thymoquinone is the potential phytochemical present in kalonji and possess strong antioxidant activity. Further, it could be used in pharmaceutical possessions to improve health standards.
