Germplasm selection and breeding by in vitro culture of wild grown Azorean blueberry (Vaccinium cylindraceum) at São Miguel island

This work aimed the in vitro establishment and micropropagation of wild Vaccinium cylindraceum shrubs selected by their fruit quality and yield characteristics. With this purpose a 2% 8-hydroxyquinoline sulfate (8-HQS) forcing solution was used to obtain explants and cultures were initiated on a Zimmerman and Broome (Z2) medium supplemented with 24.6 µM N6-[2-isopentyl] adenine (2-iP) or 9.12 µM Zeatin. The cultures proliferate in the same medium supplemented with 24.6 µM 2-iP and the obtained shoots were used to test the effect of increasing concentrations (0, 0.49, 2.46, 4.92, 9.84, and 19.68 µM) of indole butyric acid (IBA) on in vitro rooting. The effects of different substrata compositions and fertilizers (perlite, volcanic scoria, BVB-NPK, Peat-Multicote, Peat-Z2 fertigation) on plantlets development were also evaluated. The 8-HQS treatment allowed the clones’ establishment. Both 2iP and Zeatin were able to induce shoot growth in the initiation stage. Subculturing significantly increased the number of shoots per explant and the number of nodes per shoot. Best root development was achieved with 9.84 µM of IBA and plant growth was significantly improved by BVB(NPK):Perlite (1:1) in the acclimatization stage.
