An improved and effective protocol for monoconidial isolation of Pyricularia oryzae

In the present study, a protocol for monoconidial isolation of Pyricularia oryzae from infected field samples has been illustrated. This technique is based on tapping bunch of conidia on 3% water agar medium and then transferring a single germinating conidium in fresh Potato Dextrose Agar (PDA) medium following 24 hours of incubation. This protocol allows to easily locate single conidial sites, greatly shortened overall time per isolation and remarkably lowered contamination rate during isolation. Moreover, it is more convenient and effective method for the monoconidial isolation of Pyricularia oryzae that can be exploited in less funded laboratories working with this pathogen.
