Application of inverse phase gas chromatography for determination of bound water in collagen

Inverse phase gas chromatography, a technique normally used to separate and identify compounds in complex mixtures, was used to measure water sorption by muscle proteins. This procedure was found to be suitable for examining sorption processes at very low moisture content levels, such as in calculation of the moisture sorption isotherm of calf-hide collagen at 55 degrees C. The data indicated strong interactions between water and the collagen matrix at such low water vapor pressures. The new technique permitted quantitative definitions of these interactions, where data had been previously unavailable due to technical difficulties. Applications of inverse phase gas chromatography include the study of very low moisture, crystalline substances such as sucrose, where caking is a consideration in its manufacture.
