Morphological, biochemical and molecular characterization of different isolates of Fusarium from foot rot infected rice plants

Foot rot of rice caused by Fusarium verticillioides (Fusarium moniliforme) has become a major problem on certain cultivars of rice under Punjab conditions. Different types of symptoms (elongation, stunting, or both and death of plants) can be seen in nursery as well as in field. Isolations were done from foot rot infected plants showing different type of symptoms from different locations in Punjab. Total twenty isolates of Fusarium moniliforme were obtained and they were characterized on the basis of morphological characters, type of symptoms produced, production of Gibberellic and Fusaric and also by PCR using species specific primers. All the isolates fulfilled the Koch's postulates on susceptible variety under pot house conditions. Morphological identification was further confirmed by using primers specific to Fusarium moniliforme, Fusarium verticillioides and Fusarium proliferatum, Fusarium subglutinans and the primer specific for production of fumonisins were also tried. Only two primers specific for F verticillioides (VERTf1 and VertR; Vert1 and Vert2) gave amplification of the DNA. DNA of none of the isolate gave amplification with the primer pair VERTF1 & VERTF2, specific for the production of fumonisins. Detection of the Fusaric acid and Gibberellic acid was done by thin layer chromatography and by colorimetric method. All the isolates were positive for the production of Gibberellic acid and it was maximum by the isolate FR2. Out of twenty, ten isolates were positive for the production of Fusaric acid and isolate FRD produced maximum.
