The commonly used method for estimation of leucocyte ascorbic acid content described by Denson & Bowers has been criticized for ignoring the ascorbic acid content of platelets in the buffy layer. During the course of a nutrition survey of healthy young men on an Antarctic base, it was noted that leucocyte ascorbic acid estimations, using the Denson & Bowers method, were affected by the leucocyte counts measured during analysis. The effect was significantly less when leucocyte levels were at or near saturation than when subsaturated, suggesting that leucocytes share available ascorbic acid. The assessment of nutritional status from leucocyte ascorbic acid level should take account of the leucocyte count and the plasma ascorbic acid titre.
