The regulation of proline accumulation in detached leaves of rice (Oryza sativa cv. Taichung Native 1) was investigated. Increasing concentrations of NaCl from 50 to 200 mM progressively increased proline content in detached rice leaves. NaCl induced proline accumulation was mainly due to the effect of both Na+ and Cl- ions. Proline accumulation caused by NaCl was related to protein proteolysis, an increase in ornithine-delta-aminotransferase activity, a decrease in proline dehydrogenase activity, a decrease in proline utilisation, and an increase in the content of the precursors of proline biosynthesis, ornithine and arginine. Results also show that proline accumulation caused by NaCl was associated with ammonium ion accumulation.
