Effect of excess ascorbic acid in rats: Hemolysis and lipid peroxidation

The effect of oral and intraperitoneal megadoses of ascorbic acid on tissue peroxidation and hemolysis were studied. Groups of albino rats maintained on pellets were given orally 150 mg/100 g body weight of L-ascorbic acid in 1 ml saline or saline only for 28 days. Another group was given 100 mg/100 g body weight of ascorbic acid injected intraperitoneally on alternate days for two weeks. No significant weight change, mortality, or toxic symptoms were noted after the megadoses. The ascorbic acid content of tissues, except the brain, increased significantly. The in vitro lipid peroxidation of liver, brain, kidney and spleen homogenates and their catalytic efficiencies for peroxidation of linoleic acid, were depressed in the rats given ascorbic acid. The erythrocytes of the injected rats were susceptible to hemolysis in the presence of H202; the oral dose did not produce this effect.
