High-resolution chromosome preparations for G- and R-banding in Gallus domesticus

A technique used to obtain high-resolution chromosome preparations was applied to domestic fowl (Gallus domesticus). Primary fibroblast cell lines from 9-day-old chicken embryos were harvested after a double thymidine synchronization. Prometaphases and metaphases were investigated by G-banding obtained after trypsin digestion and Giemsa staining and by R-banding obtained after BrdU incorporation and Giemsa staining. As an example, the first 10 pairs of autosomes and sex chromosomes are described at a high degree of resolution, corresponding to 182 R-bands and 209 G-bands for one haploid karyotype.
