Evaluation of diclofenac toxicity with particular reference to haemato-biochemical and histopathlogical alterations in poultry

Present study was undertaken to evaluate the toxic effect of diclofenac on hemato-biochemical and histo-cellular architecture in White Leghorn (WLH) broilers. Seven weeks old female broilers (n=5) were administered diclofenac sodium @ 2.5mg/kg intramuscularly (Group-II) once daily for five days while group-I birds (n=5) served as control. Birds of group-II were found to be dull and depressed within 48 hours and had difficulty in walking as exhibited by typical hock sitting posture. There was significant (P<0.05–0.001) increase in TEC, TLC, PCV, urea nitrogen and uric acid values along with ALT and AST activities in birds of group II. On gross examination, thoracic and abdominal cavities of diclofenac-treated group birds revealed extensive deposition of white crystals on heart, liver, kidneys, lungs, spleen and joints. Histopathological examination revealed dilatation of central vein and hepatic sinusoids along with cloudy swelling in liver, infiltration of reticulo-endothelial cells and degenerative lymphoid follicles in spleen and shrinkage of glomeruli along with infiltration of leukocytes in intertubular spaces of kidneys. Findings of present study confirm the hepatotoxic, nephrotoxic and visceral gout inducing potential of diclofenac in birds.
