Vitamin C stabilizes ferritin: new insights into iron-ascorbate interactions

Cell culture technique was used to investigate ascorbate modified iron metabolism. At 1.8 nmol per ten million K562 erythroleukemic cells (equivalent to approximately 10 ug/ml), ascorbate stimulated iron incorporated into ferritin. Lysosme cells cultured for 20 hours with iron and no ascorbate showed 21% of the incorporated 59Fe in the pellet; those treated with ascorbate after loading had only 14% in the pellet. Ascorbate added to cells before pulse-labeling with 35S-methionine showed no greater incorporation of methionine into apoferitin. Finally, it was noted that cells co-cultured with ascorbate and desferrioxamine showed fewer number transferrin receptors on their surface membranes. It is concluded that the role of ascorbate in iron metabolism is related not only to enhanced absorption but also to intracellular metabolism of iron-binding proteins. (RAH)
