Elsinoe ampelina causing Paulownia scab in China

In 2019 to 2021, a scab disease of Paulownia fortunei (Seem.) Hemsl was observed in China. In the scab examination under a microscope, the conidium morphology was similar to that of Sphaceloma. Further isolation of the scab tissue was performed. Eight isolates were obtained from a host medium successfully (isolation frequency: 66.67%). Three representative isolates (EIP-1, EIP-2, and EIP-3) were used for further study. The isolates were amplified by three loci (internal transcribed spacer, RNA polymerase II second largest subunit gene, and translation elongation factor 1-α gene), and a multi-locus phylogenetic tree was built on the basis of the concatenated sequences. The results showed that the three isolates were most closely clustered with Elsinoe ampelina (de Bary) Shear (type strain CBS 208.25). E. ampelina and S. ampelina are synonyms. Thus, the isolates were identified as E. ampelina. Moreover, the pathogenicity tests confirmed that the three isolates induced evident symptoms on P. fortunei, which were consistent with those originally observed in the field.
