AFLP analysis on the genetic diversity of two hundred accessions of mango in China

200 accessions of mango (Mangifera) from foreign and domestic were used as tested materials; genomic DNA of them was extracted using the cetyltrimethyl ammonium bromide (CTAB) method from tender leaves. Eight primer combinations that produced clearly and highly polymorphic bands were screened from 64 EcoRI/MseI primer combinations. 8 primer combinations generated 1568 bands, 1539 polymorphic bands, the PPB (percentage of polymorphic bands) was 98.2%, the highest primer combination E-AAG/M-CTG was 99.0%. Genetic similarity coefficient (SC) among all germplasms varied from 0.480 to 0.838; their genetic variation varied largely. 200 accessions of mango were divided into 17 types by SC=0.665,169 germplasms clustered together at the first type. By the SC=0.720 the first type is divided into 23 sub-types, 'Yellow Mango No.1' and 'Edward No.1' has the closest relationship (SC=0.838), the two leaves, flowers, fruit shape similar, they may be the same material; 'Harris' and 'India seedling No.1' has the farthest relationship (SC=0.480). The result of cluster showed that germplasms' cluster was not completely related to their geographical origin and the type of embryo, AFLP analysis needs to combine with a variety of other molecular markers as well as the traditional classification for a comprehensive consideration.
