In vitro propagation of Rosa hybrida 'Golden Fairy' through nodal explants

In vitro propagation of rose is an essential component of rose breeding, which is a valuable economic enterprise that has of late received high importance globally. Against this back drop the present study was conducted to evaluate the in vitro regeneration potential of the hybrid tea cultivar 'Golden Fairy'. Single-node explants were cultured on Murashige and Skoog (MS) medium supplemented with various concentrations of benzyladenine (BA) and/or adenine sulphate (ADS). Out of the five treatments tried, BA singly at 2.0 mg L-1 was the best for initial inoculation based on frequency of regeneration at four weeks stage. Shoot proliferation trials were attempted on MS based medium supplemented with different concentrations and combinations of BA, IAA and ADS. Out of the four treatments employed, the highest multiple shoot regeneration was obtained on MS medium supplemented with BA (2.0 mg L-1), IAA (0.25 mg L-1) and ADS (25 mg L-1) at four weeks stage. Trials were conducted in the second subculture medium to identify the best combination of growth regulators for shoot elongation. Shoots were readily rooted on half-strength MS medium supplemented with IBA (0.25 mg L-1), activated charcoal (5%) and sucrose (2%). Initiation of rooting was observed after 2-3 weeks. After 6-7 weeks rooted plantlets were deflasked, washed and planted in potrays into a mixture of compost:cocopeat:sand 1:1:1 (v/v). Acclimatization took three weeks in a humidity chamber. Potrays with established seedlings were successfully transferred to the greenhouse.
