Growth, lipopeptide production and biocontrol efficacy against Botrytis cinerea and Monilinia fructicola of Bacillus amyloliquefaciens BUZ-14 cultured in low-cost media

The development of an economical culture media for the production of biocontrol agents (BCAs) is an essential step for their commercial use. In this study, the mass production and the biocontrol efficacy of B. amyloliquefaciens BUZ-14 cultured in media based on low cost nitrogen and carbon sources such as wheat and soy flour, starch, sugars and yeast extract have been evaluated and compared to those obtained in MOLP, an optimum medium for lipopeptides production, and in 863 medium, a control one. The supernatant antifungal efficacy was tested in vitro against Monilinia fructicola and Botrytis cinerea. Also, the production of lipopeptides, quantified through RP-HPLC, after incubation times of 24, 48, 72, 96 and 120 h was determined. In all the low-cost media, BUZ-14 reached 10(9) CFU mL(‑1), but only the supernatant obtained from 863 and MOLP was able to reduce the in vitro growth of both pathogens. Iturin A was the active antifungal lipopeptide responsible of inhibitions and both 863 and MOLP reached the highest concentration after 96 h of incubation (960 and 1550 ppm, respectively). The 863 medium is very simple in comparison to MOLP as it contains only three ingredients (peptone, glucose and yeast extract), so the formulation of a new culture medium similar to 863 with low-cost products will be the aim of further investigations for a large-scale production.
