Показать больше [+] Меньше [-]Английский. Triacylglycerols (TAGs) make up the major part of naturally occurring fats and oils. The composition and fine structure of TAGs determine to a large extent the functionality of fats and oils as food ingredients and the physiological effects of fats and oils as components of the human diet.Reversed-phase C18 high-performance liquid chromatography (RP-HPLC) is the method of choice for the separation of molecular species of TAGs. It provides a good resolution of individual TAG molecules. Thus, TAG molecules are separated according to both chain length and degree of unsaturation of the fatty acids (FAs). The complexity of natural TAG mixtures would, however, require gradient elution to fully utilize the resolution power of the HPLC system. In this chapter, we describe two methods, one for the determination of profiles of fats and vegetable oils and the second one for milk products. Both of them are performed using a binary gradient of acetonitrile (ACN)-chloroform and an evaporative light scattering detection. Indeed, the method can easily be adjusted for use with different oils by a simple modification of the gradient and run time.
