Efecto de la hemólisis de la muestra de sangre canina en el resultado de algunos analitos enzimáticos | Effect of canine blood samples hemolysis in the result of some enzyme analyses

RESUMEN: Antecedentes: la hemólisis es el proceso de destrucción de los hematíes, que conlleva a la liberación del contenido intraeritrocitario en el plasma alterando su composición. La principal molécula intraeritrocitaria es la hemoglobina, que tiene un espectro de absorción característico del grupo Hem, con un pico de 405 nm y varios picos entre 500-600 nm, lo que produce un color rojizo en el plasma proporcional a la hemoglobina liberada (Figura 1). Se suele definir la hemólisis como la aparición en el plasma de 0,3g/l de hemoglobina, que se considera la concentración mínima detectable visualmente. La hemólisis en las muestras para analizar puede interferir con los resultados, por tanto, si es una prueba mal tomada debe ser una causa de rechazo. En el laboratorio de clínica veterinaria no se tiene un estudio del comportamiento de los distintos analítos, por lo que la importancia de este trabajo se basa en determinar los efectos de la hemólisis en suero canino sobre la lectura de algunos analítos que se solicitan frecuentemente en el laboratorio. Objetivo: describir la variación que existe en los resultados de algunos analítos con respecto a la cantidad de hemólisis presente en una muestra. Método: el hemograma se realizó por la lectura en el equipo Abacus Junior Vet®; para los analítos se usó el método de Turbidimetría en el equipo A15 BioSystems QuímicaClínica. La hemólisis se indujo mecánicamente en diferentes niveles. Resultados: se empleó el análisis de varianza con un diseño completamente aleatorizado y se determinaron entre los tratamientos evaluados diferencias estadísticas a niveles de p<0,05; se realizaron además interferogramas que permiten definir el grado de interferencia si lo hubiese.Conclusión: la hemólisis en el suero canino tanto leve (0,10 - 1,00 g/l) como marcada (2,51 - 4,5 g/l) interfiere con la lectura de los analítos ALT, la Creatinina y la Urea.

ABSTRACT: Background: Hemolysis is the destruction process of red blood cells, which involves the release of the intra-erythrocytic content into the plasma altering its composition. The main intra-erythrocytic molecule is hemoglobin, that has a characteristic absorption spectrum of the heme group, with a peak at 405 nm and several peaks between 500-600 nm, and produces a reddish color in plasma, proportional to the hemoglobin released (Figure 1). Hemolysis is usually defined as the appearance of 0.3 g/l of hemoglobin in plasma which is considered the minimum visually detectable concentration. Hemolysis in the samples to be analyzed can interfere with the results and therefore, if it is a bad test taking it should be a cause for rejection. The Veterinary Clinic Laboratory does not have a study of the behavior of different analytes, so the importance of the present study is based on determining the effects of hemolysis in canine serum on the reading of some analytes that are frequently requested in the laboratory. Objective: To describe the range of variation of the results of some analytes with respect to the amount of hemolysis in a sample. Method: The blood count was performed in the Abacus Junior Vet® equipment; the Turbidimetry method in the BioSystems A15 for Clinical Chemistry was used for the analytes. Hemolysis was induced mechanically. Results: Analysis of variance with a completely randomized design was used and statistical differences at p<0.05 levels were determined. Interferograms which allow defining the degree of interference, if any, were carried out. Conclusion: Hemolysis in canine serum either mild (0.10 - 1.00 g/l) or severe (2.51 to 4.5 g/l) interferes with the reading of analytes ALT (Alanine Aminotransferase), Creatinine and Urea.
