Efeito do processamento na concentração do fator de crescimento TGF beta em lácteos | Effect of processing in the concentration of TGF beta growth factor in milk

Este trabalho teve como objetivos desenvolver um concentrado de proteínas lácteas em pó com alta concentração do fator de crescimento TGF-β e verificar como o processamento afeta a concentração do TGF-β, bem como analisar a interação que ocorre entre o TGF-β e as proteínas presentes no leite. Primeiramente, procedeu-se ao desenvolvimento do concentrado proteico, empregando técnicas de membrana (microfiltração) e dois tipos de tratamento térmico (pasteurização lenta e pasteurização rápida), e etapas de concentração a vácuo e secagem em spray dryer foram aplicadas. Foi utilizado no processamento leite cru integral proveniente de duas fontes distintas, sendo o primeiro deles obtido no estabulo da UFV e o segundo obtido no tanque de refrigeração do laticínios escola da UFV. A cada etapa do processamento foram retiradas amostras para verificar a composição centesimal e quantificar o TGF-β presente. Além disso, análise de calorimetria de titulação isotérmica (ITC) foi empregada para avaliar a interação que ocorre entre o TGF-β e as proteínas do soro e caseínas. A composição centesimal das amostras (proteínas, cinzas, extrato seco, gordura e lactose) estava dentro do previsto pelos regulamentos técnicos de identidade e qualidade (IN-62 e RTIQ – soro de leite). Os valores encontrados para o teor de TGF-β presente no leite cru está dentro do descrito na literatura, mas houve diferença na concentração do TGF-β nos leites crus empregados, sendo que o leite cru refrigerado (23,60 ng∙mL-1) apresentou valores maiores em relação ao leite cru recém ordenhado (17,43 ng∙mL-1), possivelmente pela ativação do TGF-β pela plasmina e enzimas proteolíticas presentes no leite. O TGF-β não é degradado pela pasteurização, no entanto a pasteurização rápida foi mais eficiente em recuperar esse componente em relação à pasteurização lenta, devido a maior degradação dos componentes que ocorrem nesta o que afeta a concentração do TGF-β. A pasteurização afeta a distribuição do TGF-β nas frações de proteínas do soro e caseína, o que pode ser observado na redução do teor de TGF-β2 no soro quando foi aplicado um tratamento térmico. Diferentemente de quando se aplicou a microfiltração e o TGF-β2 distribuiu-se igualmente entre a caseína e o soro. A concentração por evaporação a vácuo apresentou fator de concentração menor que 1 na concentração do TGF-β, pois observou-se maior concentração dos demais constituintes em relação ao TGF-β. Diferentemente da secagem em spray dryer que apresentou um fator de concentração maior que 1, mostrando que TGF-β2 é concentrado em maior quantidade que os demais constituintes sólidos das amostras. O uso das técnicas de membrana foi mais eficiente em concentrar o TGF-β2 tanto para o soro quanto para a caseína, quando comparado com as frações obtidas com o uso de tratamento térmico. A caseína micelar em pó (253,07 ng∙mL -1) foi o concentrado proteico que obteve maiores valores para a concentração do TGF-β2 nas condições estudadas, seguido pelo leite em pó (132,60 ng∙mL-1). TGF-β2 apresentou forte correlação (0,95) com o teor de proteína presente nas amostras. Na análise de ITC observou-se que a interação do TGF-β com as proteínas presentes no soro é exotérmica, além disso, TGF-β2 interagiu preferencialmente com as caseínas em relação as proteínas do soro, pois foi observado valores maiores de energia na primeira injeção.

This study aimed to develop a concentrated milk protein powder enriched with TGF-β growth factor and verify how processing affects the concentration of TGF-β, as well as analyzing the interaction that occurs between TGF-β and the proteins in milk. First, it proceeded to the development of the protein concentrate using membrane techniques (microfiltration) and two types of thermal treatment (slow pasteurization and pasteurization), and vacuum concentration and spray drying step were applied. It was used in processing raw milk from two distinct sources, the first obtained in the stables of UFV and the second obtained in the refrigeration tank dairy school UFV. Every step of processing samples was taken to check the chemical composition and quantify the TGF-β. Also, calorimetric isothermal titration was employed to evaluate the interaction that occurs between TGF-β and whey proteins and caseins. The chemical composition of the samples (protein, ash, dry matter, fat and lactose) was within the expected technical regulations for identity and quality (IN-62 and RTIQ – whey). The values found for TGF-β content present in the raw milk are described in the literature, but difference was observed in TGF-β concentration in raw milk employed, and the refrigerated raw milk (23.60 ng∙mL -1) showed higher values compared to raw milk recently milked (17.43 ng∙mL-1), possibly by TGF-β activation by plasmin and proteolytic enzymes present in milk. TGF-β is not degraded by pasteurization, however the pasteurization was more efficient in recovering this component in relation to slow pasteurization due to higher degradation of this component occurring which affects the concentration of TGF-β. Pasteurization affects the distribution of TGF-β in the whey proteins and casein fractions which can be observed in the reduction of TGF-β2 content in whey when a heat treatment was applied. Differently from when applied to microfiltration and TGF-β2 was equally distributed between the casein and whey. The concentration by vacuum evaporation presented lower ratio to 1 in the concentration of TGF-β, as there was higher concentration of other constituents in relation to the TGF-β. Differently form the spray drying showed that a ratio greater than 1, showing that TGF-β2 is concentrated in greater amounts that the other solid constituents of the sample. The use of membrane techniques was more effective to concentrate TGF-β2 for both, whey and casein, as compared with the fractions obtained with the use of heat treatment. The micellar casein powder (253.07 ng∙mL-1) was the protein concentrate that had higher values for the concentration of TGF-β2 under the conditions studied, followed by milk powder (132.60 ng∙mL-1). TGF-β2 showed a strong correlation (0.95) with the protein present in the samples. In ITC analysis showed that the interaction of TGF-β with the proteins present in whey is exothermic, furthermore, TGF-β2 preferably interacted with casein in relation to the whey proteins, it was observed higher energy values in the first injection.
