Implication de la toxine beta2 de Clostridium perfringens dans le syndrome de l’entérotoxémie bovine. | The beta2 toxin of Clostridium perfringens: its place in the cattle enterotoxaemia syndrome.

Английский. Cattle enterotoxaemia is an acute to hyperacute syndrome, which is characterized by the sudden death of the cattle and the presence of hemorrhagic lesions in the small intestine. Under the stimulus of misunderstood triggering factors, the enteroxaemia is associated with an exponential overgrowth of Clostridium perfringens in, often limited, small intestine segments and the production of toxins acting locally or systemically. Among the numerous potentially implicated toxins in the genesis of these lesions, the beta2 toxin (CPB2) deserves special attention. Discovered in piglets suffering from necrotic enteritis (Gibert et al., 1997), it was thereafter detected in other animal species suffering of various diseases, but also in healthy animals. Two alleles were quickly defined (1) consensus cpb2, associated to swines’ strains able to produce CPB2 and atypical cpb2, detected in strains from other species and a priori unable to express CPB2 (Jost et al., 2005). The strains that could produce CPB2 were widely distributed but the real implication of this toxin in intestinal diseases is not yet clarified. In cattle, the results of a study on intestinal ligated loops in our lab, suggested a synergistic effect of alpha and beta2 toxins to explain the hemorrhagic lesions observed in this assay (Manteca et al., 2000). The present work, begun in 2000, is the continuation of this study. The global goal was to determine wether the CPB2 toxin was associated, or not, with the cattle enterotoxaemia observed in field conditions. Prior to realizing a case/ control study, we had to produce and purify a CPB2 toxin from cattle strains and we had to have specific tools able to detect the cpb2 gene and the corresponding toxin. The whole work was organized in 6 studies. The 3 first studies served to create new and adapted tools to detect the cpb2 gene and the corresponding CPB2 toxin from cattle C. perfringens strains. The 3 last studies investigated the prevalence of cpb2 gene and ability of the cattle strains to produce CPB2 toxin in vitro in conventional and veal calves farms.

Французский. L’entérotoxémie bovine est un syndrome aigu à suraigu caractérisé par une mort subite et des lésions nécrohémorragiques au niveau de l’intestin grêle. Sous l’influence de facteurs déclenchants encore mal connus, l’entérotoxémie est associée à une multiplication exponentielle de Clostridium perfringens dans des portions souvent limitées de l’intestin grêle et une production de toxines à actions locales et systémiques. Parmi les nombreuses toxines potentiellement impliquées dans la genèse des lésions, la toxine beta2 (CPB2) mérite une attention particulière. Elle a été découverte chez des porcelets atteints d’entérite nécrotique (Gibert et al., 1997). Par la suite, de nombreuses études ont permis de la mettre en évidence chez d’autres espèces animales dans des circonstances pathologiques diverses ainsi que chez des animaux sains. Deux allèles du gène cpb2 ont rapidement été définis : cpb2 consensus, associé initialement aux souches porcines, capables d’exprimer CPB2 et cpb2 atypiques, mis en évidence dans les autres espèces et a priori incapable d’exprimer la toxine (Jost et al. 2005). Les souches qui produisent la CPB2 sont, donc, largement distribuées, mais l’implication de cette toxine dans les pathologies intestinales n’est pas encore élucidée. Chez les bovins, les résultats d’une étude réalisée dans notre laboratoire sur anses intestinales ligaturées ont suggéré une action synergique entre la toxine alpha et la toxine CPB2 pour expliquer les lésions hémorragiques observées dans le modèle (Manteca et al., 2002). C’est dans le prolongement de cette étude que le présent travail a débuté en 2000. L’objectif général était de déterminer si la toxine CPB2 est ou non associée à des cas d’entérotoxémie bovine rencontrés sur le terrain. Avant de réaliser ces enquêtes cas/témoins, il a fallu au préalable produire une toxine CPB2 d’origine bovine, la purifier et se doter d’outils de détection spécifiques du gène cpb2 et de la toxine. L’ensemble du travail a été organisé en 6 études. Les 3 premières études visaient à mettre au point les bons outils pour détecter le gène cpb2 et la toxine correspondantes chez des souches de C. perfringens bovines. Les 3 dernières investigaient la présence du gène cpb2 et la capacité d'expression de la toxine CPB2 in vitro par des souches provenant de veaux d'élevage conventionnel ou d'étable d'engraissement.
