X-ray crystallographic studies on two cysteine-rich anticarcinogenic microproteins | Röntgenkristallographische Untersuchungen an zwei cysteinreichen antikarzinogenen Mikroproteinen

Zwei cysteinreiche, antikarzinogene Mikroproteine wurden mit röntgenkristallographischen Methoden untersucht.Der aus den Limabohnen isolierte Trypsin-Chymotrypsin Inhibitor (LBTI) gehört zu der Bowman-Birk-Familie der Serin-Protease-Inhibitoren. Die ungewöhnlich hohe Anzahl der Schwefelatome und die hohe Symmetrie der Raumgruppe ermöglichten die Strukturlösung anhand der in-house gemessenen anomalen Streuung von Schwefel. Die LBTI-Moleküle nehmen die der Bowman-Birk-Familie entsprechende bow-tie- >Faltung an. Sie bilden in der Kristallstruktur Dimere derart, dass die reaktiven Schlaufen unabgeschirmt bleiben.Der ternäre Komplex des LBTI mit dessen beiden Zielenzymen konnte mit der Methode des molekularen Ersatzes problemlos gelöst werden. Diese Struktur erhellte die wichtigsten Faktoren, die die Enzymspezifität beeinflussen können, wie die asymmetrische Verteilung der hydrophoben und hydrophilen Oberflächen an der Enzym-Inhibitor-Schnittstelle, oder Konformationsänderungen während einer gleichzeitigen Enzymbindung.Viscotoxine (VT) sind Mitglieder der Alpha- und Beta-Thionin-Familie und besitzen eine cytotoxische Wirkung, indem sie biologische Membrane zerstören können. Die Kristallstruktur der VT A3 Isoform wurde ebenfalls mit Hilfe der anomalen Dispersion des Schwefels gelöst. In diesem Fall verlangte die relativ niedrige Auflösung des anomalen Signals eine Verbesserung des Schweratom-Suchverfahrens für eine erfolgreiche Phasenbestimmung. Die dreidimensionale Struktur der VT A3 Moleküle ähnelt dem Buchstaben "L". Eine positiv geladene Bindungstasche konnte zwischen dem kurzen und langen Schenkel der L-Form lokalisiert werden, wo ein funktionell bedeutsames Phosphation gebunden ist. Die VT A3 Moleküle liegen in der Kristallstruktur als Dimere vor, die weiter zu Tetrameren verbunden sind.Die ausgezeichneten Diffraktionseigenschaften der Kristalle aus der Isoform B2 ermöglichten die Datensammlung bis zu atomarer Auflösung und die Strukturlösung mit ab initio direkten Methoden. Die in der relativen Lage der Monomere beobachteten Unterschiede und Sequenzvergleich der Isoformen deuten darauf hin, dass die für eine hohe cytotoxische Aktivität verantwortlichen hydrophoben Aminosäurereste an der äußeren Oberfläche des längeren L-Schenkels liegen.

Two cysteine-rich, anticarcinogenic microproteins belonging to distinct protein classes were studied with X-ray crystallographic methods.The lima bean trypsin-chymotrypsin inhibitor (LBTI) can be classified into the Bowman-Birk serine proteinase family. The unusually high cysteine content and the high symmetry space group enabled the structure solution of the free inhibitor by means of the anomalous signal of sulfur atoms measured in-house. The LBTI structure resembles the bow-tie fold canonical in the Bowman-Birk family. The inhibitor molecules associate to dimers in the crystal structure in such a manner that the reactive site loops remain unshielded.The ternary complex of LBTI with its both target enzymes could be easily solved by molecular replacement. The structure shed light on important factors that can influence the enzyme preference of inhibitors in the Bowman-Birk family, such as the asymmetric distribution of hydrophobic and hydrophilic patches on the enzyme-inhibitor interface and conformational changes upon simultaneous enzyme binding.Viscotoxins (VTs) belong to the family of alpha- and beta-thionins that exert cytotoxic activity and are able to disrupt biological membranes. The crystal structure of the VT isoform A3 was also solved exploiting the weak anomalous signal of sulfur atoms measured in-house. In this case the medium resolution anomalous signal required an improvement in disulfide location for successful phasing. VT A3 molecules adopt the canonical ?- and ?-thionin fold that is similar to a capital letter L. A positively charged binding pocked was located between the two branches of the L structure, where a functionally essential phosphate ion is bound. Molecules of the asymmetric unit were observed to form dimers and further associate to tetramers.Favourable diffraction properties of crystals of the VT isoform B2 enabled data collection to atomic resolution and structure solution with ab initio direct methods. Differences in the relative position of the monomers in the dimer and sequence alignment analysis suggest that hydrophobic residues responsible for high cytotoxic activity are located on the external surface of the longer arm of the letter L.
