Untersuchung zum Einfluss des Lipidgehaltes in Oozyten des Rindes auf die embryonale Entwicklungsfahigkeit nach In-vitro-Maturation und In-vitro- Fertilisation | Research on the bovine oocyte's lipid content and its influence in embryonic developmental capacity before and after in vitro maturation and in vitro fertilization

Trotz zahlreicher Verbesserungen in den IVP-Protokollen beträgt der Prozentsatz der in vitro produzierten, transfertauglichen Blastozysten zwischen 30-45%. Das Ausgangsmaterial in Form der Oozyten unterschiedlicher Entwicklungs- und Reifestadien aus den Ovarien von Kühen unbekannten Alters und Gesundheitszustandes waren heterogen, ebenso wie die Entwicklungskompetenz der gewonnenen Oozyten. Neben Eizelldurchmesser, Kernstatus, Cumulusausstattung und Beschaffenheit des Zytoplasmas ist der Lipidgehalt in Oozyten und Embryonen ein wichtiger Parameter für ihre Qualität und Kryotoleranz. Der Lipidgehalt in Oozyten korreliert mit der morphologischen Erscheinung des Zytoplasmas. Gegenstand der Untersuchung dieser Arbeit war die Bestimmung des Lipidgehaltes in bovinen Einzeloozyten der Rasse Deutsches Fleckvieh hinsichtlich der Intensität und des Lipidmusters im Ooplasma sowie der parallelen Charakterisierung der Chromatinkonfiguration in Abhängigkeit von verschiedenen Cumulus-Oozyten- Komplex-Klassen (COK-Klassen) vor und nach der In-vitro-Maturation (IVM). Die Beurteilung der weiteren Entwicklungsfähigkeit erfolgte anhand von IVF und IVC. Basierend auf der Messung der Fluoreszenzintensität des Zytoplasmas zeigten die Oozyten der Klassen I, II und III ähnliche Werte (638 ± 34; 575 ± 43 und 715 ± 60 µA/ Oozyte), waren aber signifikant höher als in den Oozyten der COK-Klasse IV (326 ± 45 µA/ Oozyte). Nach der IVM war die Fluoreszenzintensität und damit der Lipidgehalt in den Oozyten der Klassen I und II höher (736 ± 39 und 631 ± 28 µA/ Oozyte), in der Klasse III (558 ± 49 µA/ Oozyte) hingegen war eine signifikante Abnahme zu beobachten. Es wurden drei wiederkehrende Lipidmuster festgestellt, die abhängig von der COK-Klasse und dem Entwicklungsstadium der Oozyte auftraten. Die vor der Maturation vorherrschende feinkörnige Zytoplasmastruktur veränderte sich im Verlauf der Reifung zu einer flockigen Struktur. Die Entwicklungsraten zur Blastozyste am Tag 8 nach IVF war in den Klassen I und II signifikant höher (32,7% und 30%) als in den Gruppen III und IV (5,5% und 0%). Im zweiten Teil der Arbeit wurden zwei unterschiedliche Seren als Medienzusatz untersucht. Der Lipidgehalt in den Oozyten wurde durch das verwendete Serum beeinflusst. Die höchste Fluoreszenzintensität konnte in den Oozyten nach IVM in ECS-supplementiertem Medium gemessen werden. Unterschiede im Lipidmuster waren hier ebenfalls zu beobachten. Weiterhin führte die Mediensupplementierung mit ECS oder FCS zu Unterschieden in der frühen embryonalen Entwicklung. Nach der embryonalen Entwicklung (Tag 8 nach IVF) war der Lipidgehalt signifikant höher als an Tag 2 nach IVF. Die höchste Intensität wurde bei Verwendung von ECS am Tag 8 nach IVF (799 ± 18,3 vs. 760,1 ± 22,2 µA/ Embryo) gemessen. Nicht nur der Anteil Blastozysten (31,8% vs. 24,6%), sondern auch die Anzahl der Zellkerne pro Blastozyste (103 vs. 88) stieg nach ECS-Supplementierung des Mediums signifikant an. Aus den Ergebnissen wird deutlich, dass entwicklungskompetente Oozyten (COK-KIassen I und II) Lipide aus dem umgebenden Milieu aufnehmen und sich dieser Anstieg auf die frühe Embryonalentwicklung positiv auswirkt. Unterschiedliche Serumzusätze führen zu unterschiedlicher Embryonenentwicklung. Die Verwendung von ECS als Serumzusatz zum Medium führte in allen untersuchten Parametern zu höheren Werten und zu besseren Entwicklungsraten. In weiteren Versuchen ist zu verifizieren, ob diese in vitro produzierten Embryonen einen Vorteil für die weitere Entwicklung besitzen und bessere Überlebensraten nach Übertragung in Empfängertiere zeigen.

Despite significant improvements in the ivp-protocols, the percentage of in vitro produced blastocysts suitable for transfer is 30-45%. The base material, being oocytes of variable development and maturity stages taken from the ovaries of cows of unknown ages and health status, as well as the developmental competence of these received oocytes are very heterogeneous: Besides oocyte diameter, nuclear status, cumulus morphology, and the composition of the cytoplasm, the lipid content of oocytes and embryos is an important parameter for their quality and cryotolerance. The lipid content of immature oocytes is correlated with the morphology of the cytoplasm. The purpose of this research project was to indirectly determine the lipid content of bovine oocytes of the breed German Fleckvieh through measuring their fluorescence intensity. At the same time, lipid patterns in the ooplasm were categorized and the chromatin configuration of oocytes of diverse classes of cumulus-oocyte complexes (coc-classes I-IV) were characterized before and after in vitro maturation (ivm). Evaluation of onward viability after in vitro fertilization was conducted in two parallel experimental approaches with different serum supplements. After measurement of the cytoplasmic fluorescence intensity of oocytes before IVM, oocytes from group I, II and III had similar fluorescence intensities (638 ± 34; 575 ± 43 and 715 ± 60 µA/ oocyte, respectively) but these were significant higher than those of oocytes of group IV (326 ± 45 µA/ oocyte). After IVM the fluorescence intensities in group I- and II-oocytes were increased (736 ± 39 and 631 ± 28 µA/ oocyte) and in group III significantly decreased (558 ± 49 µA/ oocyte). At the time of oocyte collection class I and II oocytes were mainly in the diplotene stage (76.5% and 83.6%, respectively) while class III oocytes had already reached later stages of meiosis (diakinesis to metaphase II). At the end of ivm the proportion of degenerated oocytes was significantly higher in class IV oocytes than in other coc-classes. Three recurring lipid patterns were detected, which occured dependent on the coc-class and the stage of development of the oocyte. The fine-grained morphology, which prevailed before maturation, changed to a fluffy appearance after maturation. The rate of development to the blastocyst stage after IVF was significantly higher in group I and II (32.4% and 30.7%) than in group III and IV (6.7% and 0%, respectively). In the second part of the study the effect of serumsupplementation (ECS vs. FCS) was investigated. The lipid content in oocytes was influenced by the application of serum (ECS, FCS) in media for IVM and IVC. The highest fluorescence intensity was determined in oocytes after IVM in ECS supplemented medium. There were also different lipid distribution patterns after IVM with different serum supplementation. Furthermore, the supplementation of media with ECS or FCS led to differences in the early embryonic development. During embryonic development the lipid content increased significantly from day 2 to day 8 using both sera. The highest intensity was measured with ECS on day 8 (799.6 ± 18.3 vs. 760.1 ± 22.2 µA/ embryo). Not only the proportion of blastocysts (31.8 vs. 24.6%) but also the number of nuclei per blastocyst (103 vs. 88) was significantly increased after maturation and culture with ECS. The results show that developmental competent oocytes (COC class I and II) are able to incorporate lipids from the culture medium. This incorporation could be the reason for the increase of the lipid content and the positive effect on development of the embryos. Different serum supplementation led to different development of embryos. ECS as serum supplementation to the medium led to higher values in all parameters and better development. The question if the produced embryos have advantages in their further development as well as better survival rates after their transfer into the recipient must be confirmed in further studies.
